0.19.0 is a spectral-data release. Every one of the 85 fluorophores that had only ever been drafted was checked against a manufacturer or curator source, and 26 of them turned out to be wrong -- including Alexa Fluor 488 and DRAQ5, whose stored emission was off by 16 nm. Those values are corrected and cited. 143 of the pack’s 157 entries now carry a source; the 14 that do not are named below, because an honest gap is more useful than a confident guess.
What 0.19.0 changes.
Alexa Fluor 488 was stored as 490/525 nm; Thermo’s own table says 495/519. DRAQ5’s emission was 16 nm low. ATTO 633 was 6 nm out. Each corrected value now carries the manufacturer page it came from, with the date it was read.
Up from 72. Seven whole probe families -- MitoTracker, LysoTracker, SYTOX, BODIPY, CellMask, LIVE/DEAD and SYTO -- had every variant sourced, so the family can finally be marked as cited rather than drafted.
Cy2, Cy7 and TRITC had no primary source worth citing; DCFDA’s source gives a range, not a peak. Nine more are ambiguous names rather than bad numbers -- see below.
CFP stores the values of ECFP; GFP stores EGFP’s; IRFP stores iRFP713’s; mRuby stores the original, not mRuby2. The numbers are real, but the key doesn’t say which variant it means. Left alone for now, because renaming a key would break saved studies.
A cited entry means its two peak values match a source we fetched and recorded. It does not mean a microscopist checked it, and the Gaussian curve widths in the spectral view remain estimates throughout.
